PhD Researcher: Unveiling the Dynamics of Ubiquitin Processing by Cdc48/p97

The Ubiquitin Processing Motor Cdc48/p97 is a pivotal enzyme in cellular regulation, controlling protein degradation, membrane remodeling, and DNA replication. As a PhD Researcher in our group, you will investigate the dynamic action of this motor using innovative (bio)physics experiments. You will work in the Tans biophysics lab at AMOLF Amsterdam, employing optical tweezers and single-molecule fluorescence to visualize the binding, unfolding, and translocation of ubiquitin molecules by Cdc48/p97. This project is part of an international team, with other groups specializing in in-vitro biology and biochemistry. Your aim is to elucidate the dynamic mechanisms underlying this pivotal function, addressing fundamental questions such as the role of ubiquitin loops and branch points, the speed, force, and pause dynamics of Cdc48/p97, and the influence of regulatory proteins on this process.

Key Responsibilities:

  • Design and perform innovative (bio)physics experiments to investigate the dynamic action of Cdc48/p97
  • Employ optical tweezers and single-molecule fluorescence to visualize the binding, unfolding, and translocation of ubiquitin molecules
  • Collaborate with international partners in Germany to obtain complementary in-vitro data
  • Analyze complex temporal data and formulate new models to explain your findings
  • Develop new experimental schemes and adapt existing biochemical protocols

Requirements:

  • Master's degree in experimental physics or chemistry

Working Conditions:

  • Full-time appointment (40 hours/week, 12 months/year) in the service of NWO-I
  • Starting salary: €2,781 gross per month
