3 jaar **_Neuroinflammatie is een inflammatoire respons van de hersenen of het myelum, die kan worden veroorzaakt door zowel externe als interne factoren. Het is een relatief nieuw concept, aangezien de hersenen tot voor kort werden gezien als 'immune-privileged': buitengesloten van het immuunsysteem door de bloed-hersenbarriere. Neuroinflammatie blijkt een grote rol te spelen bij een verscheidenheid aan neurologische symptomen en ziekten. Dit onderzoek richt zich op het in beeld brengen van neuroinflammatie met behulp van Magnetic Resonance Imaging (MRI) in Inflammatory Bowel Disease (IBD). IBD - een verzamelnaam voor onder andere de ziekte van Crohn en colitis ulcerosa - is een chronische systemische inflammatoire aandoening waarbij vooral het darmstelsel is aangedaan. Een groot deel van de patiënten met IBD heeft echter ook last van neurologische symptomen, zoals chronische vermoeidheid en concentratiestoornissen, zonder dat hier een duidelijke oorzaak voor is. Deze klachten kunnen zo heftig zijn dat de kwaliteit van leven sterk achteruit gaat. Uit eerdere studies is gebleken dat chronische vermoeidheid en concentratiestoornissen geassocieerd zijn met neuroinflammatie.

Onze hypothese is dat IBD zich niet alleen primair beperkt tot het darmstelsel, maar ook een bepaalde mate van neuroinflammatie veroorzaakt. Als dit inderdaad het geval is, zouden we op zoek moeten gaan naar behandelopties die ook de inflammatie in de hersenen aanpakken. Met behulp van MRI-scans specifiek gericht op neuroinflammatie zullen we onze hypothese gaan testen.

Het promotieonderzoek zal bestaan uit (1) een literatuuronderzoek naar neuroinflammatie in systemische inflammatoire aandoeningen (waaronder IBD); (2) een klinische pilotstudie waarin we op zoek gaan naar neuroinflammatie met een 7 tesla (7T) MRI-scanner in patiënten met IBD en gezonde vrijwilligers; en (3) opzet voor - en mogelijk start van - een grotere klinische studie waarin wordt gekeken naar de invloed van o.a. medicatie bij de ontwikkeling van neuroinflammatie in IBD.
- Voor de 7T MRI-scans moeten we gebruik maken van de scanners in Utrecht en Essen, Duitsland; dit kan - zeker wanneer de klinische studies lopen - leiden tot aanzienlijke (vergoedde) reistijd, waardoor het bezit van een auto handig is (maar geen must)._

**Taken en verantwoordelijkheden**
- Gedegen literatuuronderzoek opzetten en analyseren van de gevonden publicaties op passende wijze.
- Een klinisch wetenschappelijk onderzoek ontwerpen en uitvoeren, inclusief het schrijven van een METC-protocol, includeren van patiënten, en (meehelpen met) MRI-scans maken.
- Onderzoeksgegevens (waaronder MRI-scandata) analyseren en deze presenteren in wetenschappelijke publicaties en op wetenschappelijke bijeenkomsten.
- Samenwerken met een gevarieerd team waaronder clinici, MR-fysici, engineers en laboranten.
- Het project voltooien met een wetenschappelijk proefschrift.

Het promotietraject zal gedurende een tijdsbestek van 3 jaar plaatsvinden in voltijd; deeltijd (28,8 uur) behoort in overleg ook tot de mogelijkheden. Bij het Radboudumc bouw je aan de toekomst. - Inschaling is afhankelijk van vooropleiding. Voor een basisarts is schaal 10 van toepassing: tussen de € 3.493 en € 5.504 (fulltime). Voor overige studierichtingen geldt inschaling in schaal 10A: bij aanvraag van het dienstverband start jij in schaal 10A, stap 0 (€ 3.017 op basis van een fulltime dienstverband). In maximaal 4 jaar tijd groei je door naar schaal 10A, stap 3 (€3.824 op basis van een fulltime dienstverband). Ook krijg je 8% vakantiegeld, 8,3% eindejaarsuitkering en indien van toepassing op de functie, 47% tot 72% onregelmatigheidstoelage.
